Design: Genomic DNA was isolated from FFPE samples by the QIAamp DNA FFPE Tissue Kit (Qiagen, USA) according to manufacturers instructions. Matching genomic DNA from white blood cells (WBC) of the same individual was extracted from the buffy coat by GeneJET PCR Purification Kit (ThermoFisher Scientific) according to manufacturers instructions. DNA fragmentation and library preparation for paired FFPE and WBC samples were performed using the NEBNext Ultra II FS DNA library prep kit (New England Biolabs, USA) following the manufacturers instructions. Libraries were pooled together and hybridized with predesigned probes for 473 targeted genes (Integrated DNA Technologies, USA), as well as the promoter of TERT and selected intron of 6 genes frequently rearranged in cancer (NTRK1/2/3, RET, ALK, ROS1). Massive parallel sequencing of DNA libraries was performed on the DNBSEQ-G400 sequencer (MGI, China)
